I've created a custom post type and a custom taxonomy. I can create new posts and assign those taxonomies, but it returns a 404 when I go to mydomain.com/mycustomtaxonomy/termslug.

Here is my code:

I've created: taxonomy-familia_videos.php

I've refreshed the permalinks structure, but I always get a 404.

Any help?

The problem is that you're expecting behavior that simply doesn't exist. Just like mysite.com/category will 404, so will mysite.com/mycustomtaxonomy
