I have a custom post type and I want to create a page which will automatically list all the comments posted under my single-{my-post-type}.php.

What is the best practice for creating something like ?

Should I create a custom page called let's say single-my-post-type-comments.php and put there the neccesarry code ?

Is this possible in wordpress?

Creating a page and custom template for it allows you to use completely arbitrary code in that template, that seems like a sound way to go. –  Rarst Nov 5 '13 at 18:59
